Effects of chelators and calcium ions on the response of renal adenosine triphosphatase activity to hormones and concanavalin A.

چکیده
Experiments with plasma membrane from rat liver have been interpreted as showing that the activity of the (Na++K+)-dependent ATPase* is subject to modulation by the action of hormones, such as L-adrenaline (epinephrine) and insulin, through the latter's effects on adenylate cyclase activity and consequent changes in cyclic AMP concentration (Luly et af. , 1972; Tria et af . , 1974; Luly & Emmelot, 1975). Because of the obvious physiological importance of such hormonal control of the ATPase activity, if the interpretation is correct, we have examined this system further. Rat kidney tissue was used instead of liver because the former is a better source of (Na++K+)-dependent ATPase. When the same method for preparation of a membrane fraction (Ray, 1970) was applied to kidney and the (Na++K+)-dependent ATPase activity was assayed under the conditions used by Bonting (1970), results closely paralleling those of Luly & Emmelot (1975) were obtained (Tables 1 and 2). However, no such effects were observed when the (Na++K+)-dependent ATPase activity was assayed under the conditions described by Wheeler et al. (1975). Comparison of the differences between the two assay conditions revealed that the presence of 0.5mM-EGTA in place of 0.lmM-EDTA abolished the effects of the hormones and concanavalin A. The EGTA appeared to mimic the effect of concanavalin A and adrenaline. For example, an (Na+ + K+)-dependent ATPase activity (in pmol of PJmin per mg of protein) of 0.13 was decreased to 0.04 by concanavalin A, and to 0.06 by EGTA. Similarly, when Ca2+ was omitted from the solution used for the initial homogenization of the tissue, the final preparation had much lower (Na++K+)dependent ATPase activity than usual (0.07unit). Moreover, that low activity was no longer sensitive to adrenaline or concanavalin A.

Effect of Thyroid Activity Modulation on Some Histological and Biochemical Aspects in Broiler Chicks
An experiment was conducted to investigate the effect of thyroid gland activity modulation (Hyper or hypothyroidism) on energy utilization in broiler chickens. Two hundred and forty one-day-old Cobb broiler chicks were distributed into three dietary metabolizable energy (ME) treatment groups (80 chicks each). The control group (E0) was fed basal diet (3150 kcal/kg diet), the second group (E1) w...
متن کاملThe effects of high calcium intake on the activity of thyroid gland in rats
The effects of goitrogens on the thyroid gland have been much studied and calcium is considered as a goitrogen in some cases. Since the level of calcium is high in drinking water of some cities of Khozestan province, the effects of high calcium intake on the function of thyroid gland is investigated in this study. For this purpose the concentration of thyroid hormones is measured in rats. The s...
متن کاملThe Inhibition of the Adenosine Triphosphatase Activity of Actomyosin by Magnesium Ions
In 1939 Engelhardt and Ljubimova (2) suggested that myosin (the contractile substance of muscle) and adenosine triphosphatase are probably identical, and that 'the calcium ion is a specific activator of this enzyme. Working with a myosin suspension made from rabbit muscle (by the method of Greenstein and Edsall (4) or of Weber and Meyer (11)) D. M. Needham (9) tested the activating powers of bo...
متن کاملCaclium uptake and associated adenosine triphosphatase activity in fragmented sarcoplasmic reticulum. Requirement for potassium ions.
The effects of monovalent cations on calcium uptake by fragmented sarcoplasmic reticulum have been clarified. Homogenization of muscle tissue in salt-containing solutions leads to contamination of this subcellular fraction with actomyosin and mitochondrial membranes. When, in addition, inorganic cations are contributed by the microsomal suspension and in association with nucleotide triphosphate...
متن کاملEffect of alkali-treated lipopolysaccharide on the intracellular cations of human erythrocytes.
The adsorption to human erythrocytes of Escherichia coli lipopolysaccharide treated by mild alkaline hydrolysis (h-LPS) stimulated an increase in the intracellular Na+ concentration and a decrease in the intracellular K+ concentration of the erythrocytes. Erythrocytes treated by h-LPS remained responsive to the membrane adenosine triphosphatase inhibitors ouabain and ethacrynic acid, indicating...
